Abstract: 

The aim of this study was to investigate the effect of using portulaca oleracea as a source of plant antioxidants for feeding fattening male lambs, on carcass traits, meat quality, and OXIDATIVE stability. Twenty-one male lambs with an average weight of 24±1.5 kg and 150±15 days old were fed with one of the experimental rations included a diet without portulaca oleracea as control, a diet containing 7.5%, and a diet containing 15% portulaca oleracea, for 84 days. The muscle (Longissimus dorsi) samplewas used to determine pH, chemical composition, colorimetric properties, and OXIDATIVE stability of meat. Meat lipid oxidation was determined after 1, 7, and 30 days of refrigerated storage using thiobarbituric acid (TBARS). The use of portulaca oleracea in diets improved meat color characteristics such as L, chroma, and Hue. The highest ash concentration and the lowest meat moisture were observed in the treatment of 15% portulaca oleracea (P<0.05). Meat pH was the lowest in the first hour after slaughter but did not differ at 24 h. Compared to the control, in both treatment groups feeding the lambs with portulaca significantly reduced the concentration of Malondialdehyde in the meat of the lambs and increased the meat OXIDATIVE stability (P<0.05). The results of this experiment showed that feeding portulaca oleracea by improving properties such as color and increasing the meat OXIDATIVE stability improves meat quality, which may have lead to an increase in its nutritional value in terms of human health.

Abstract: 

Methyltriphenylphosphonium peroxydisulfate (MTPPP) was synthesized by treating methyltriphenylphosphonium bromide with potassium peroxydisulfate in aqueous solution. This reagent was used as a suitable and efficient reagent for the CLEAVAGE of oximes, phenylhydrazones and 2,4-dinitro phenylhydrazones to their corresponding carbonyl compounds under non-aqueous condition.

Abstract: 

Few novel cobalt (II) and copper (II) complexes [M (fmp)3] Cl2, [M (fmp) (bpy)2] Cl2, [M (fmp) (phen)2] Cl2 and [M (fmp) (phen) (bpy)] Cl2 (fmp=3-furan-2-ylmethylene-pentane-2, 4- dione, phen=1, 10-phenanthroline, bpy=2, 2' -bipyridine) have been synthesized and characterized by elemental analyses, molar conductance, magnetic susceptibility measurements, IR, electronic, EPR, mass spectra and cyclic voltammetric studies. The synthesized complexes are found to be monomeric and electrolytic nature. The UV-Vis., magnetic susceptibility and EPR spectral data of the complexes suggest a distorted octahedral geometry for Cu (II) complexes and octahedral geometry for Co (II) complexes around the central metal ion. The CV profile of the complexes shows a quasi-reversible peak which indicates that the metal-ligand linkage is more covalent in nature. Spectroscopic and viscosity measurements prove that the [M (fmp) (phen)2] Cl2 complexes bind more efficiently with DNA than other complexes through intercalative mode. The difference of peak potential and electrochemical parameters between free and DNA-bound complexes shows the formation of an electrochemical active complex between synthesized complexes and DNA. All the complexes cleave the Supercoiled pUC19 DNA in vitro under the presence of reducing agent (3-mercaptopropionic acid). The in vitro antimicrobial activities of the compounds have been tested against the bacterial and fungal strains using the disc diffusion method. The minimum inhibitory concentration (MIC) values against the growth of microorganisms are much larger for metal chelates than the individual ligands.

Abstract: 

Recently, we demonstrated chromosome instability (CIN) in human CLEAVAGE stage embryogenesis following in vitro fertilization (IVF). CIN not necessarily undermines normal human development (i.e. when remaining normal diploid blastomeres develop the embryo proper), however it can spark a spectrum of conditions, including loss of conception, genetic disease and genetic variation development.To study embryonic CIN further we have developed new methods based on high-resolution microarray as well as next-generation sequencing technology that characterize the genome of a single human cell. We delivered proof-of-principle for detecting various types of structural variants, including Mb- to Kb-sized duplications and deletions, in single human (tumor) cells by low coverage sequencing and mapping.Based on the copy number changes that were detected by single-cell microarray analysis of multiple blastomeres of the same embryo, it was hypothesized that chromosome breakages and fusions occur frequently in human CLEAVAGE stage embryos and instigate subsequent breakage-fusion-bridge cycles. In addition, we hypothesized that the DNA breaks present in spermatozoa could trigger this CIN. To test these hypotheses, we genotyped both parents as well as 93 blastomeres from 24 IVF embryos and developed a novel SNP-array based algorithm to determine the parental origin of (aberrant) loci in single cells. Paternal as well as maternal alleles were commonly rearranged in the blastomeres indicating that sperm-specific DNA-breaks do not explain the majority of these structural variants. In addition, single-cell genome sequencing together with parent-oforigin SNP-array and microarray-guided FISH analyses demonstrate that breakage-fusion-bridge cycles as well as more complex rearrangements are sparked in the human CLEAVAGE stage embryo.Our data provide evidence that the human CLEAVAGE stage embryo is likely an important source of constitutional chromosomal disorders. The developed single-cell genome analysis methods are generic and will deliver novel insights in embryo and tumor genome research.
